From an early age, Summer trained to be a ballerina. Besides traditional ballet, she studied tango and flamenco. She danced at the Majestic Theatre. Glau's first television credit was a guest role as a ballerina on the 2002 episode Waiting in the Wings of the television series Angel. There, she caught the eye of director Joss Whedon, who later cast her in his critically acclaimed but ultimately short-lived TV series Firefly as the much traumatized River Tam. After Firefly's cancellation, she appeared in several famous series such as Cold Case on the episode "Love Conquers Al" and in CSI on the episode "What's Eating Gilbert Grissom?". She also had a small part in the MGM feature Sleepover as a ticket girl.

In 2005, she reprised her role as River Tam in the movie Serenity. In 2006, she starred in the science fiction film Mammoth as Jack. She also appeared in the ABC Family TV movie The Initiation of Sarah (a remake of a 1978 horror film) as Lindsey Goodwin the (normal) sister of Sarah Goodwin (Mika Boorem) a young woman with mysterious powers.

In the second season of The Unit, she portrayed Crystal Burns, the girlfriend of Jeremy Erhart (Ex boyfriend Daniel Wisler). Glau also played Tess Doerner, a paranoid schizophrenic returnee in the second season premiere of The 4400 (episode "Wake-Up Call"), and became an essential recurring character from Season 3 onwards.

In Terminator: The Sarah Connor Chronicles. which debuted on January 13, 2008, Glau plays the role of Cameron Phillips, a Terminator infiltration unit sent back to protect John Connor and Sarah Connor from Skynet. Summer also guest starred as herself in episode seventeen of the second season of The Big Bang Theory. From 2009-2010, she joined the cast of Joss Whedon's Dollhouse. She portrayed Bennett Halverson, the programmer for a rival Dollhouse in episodes 5,6,11 and 13: The Public Eye, The Left Hand, Getting Closer and Epitaph 2.

Summer played Lindsey Forrest in the Lifetime movie Deadly Honeymoon that aired on 25 April 2010. Glau also provided the voice of Supergirl in the animated movie "Superman/Batman: Apocalypse which was released on September 28 on DVD, Blu-ray, On Demand and Digital Download. She also voiced the character in the video game DC Universe Online. Summer Glau was also a guest star on Chuck as Greta.

The Cape premiered on NBC during the 2010–2011 television season as a mid season replacement beginning with a 2-hour pilot episode on Sunday, January 9, 2011 at 9 PM Eastern and Pacific Time continuing on Monday evenings in the 9 pm timeslot beginning on January 17. NBC cut the total number of episodes to be aired from 13 to 10 as a result of low ratings.On March 2, 2011, NBC announced that the series finale would only be aired on the network's website. One of the people helping The Cape in his war against crime was the computer whiz Orwell played by Summer Glau.

Post-prod for late 2011 release is the western movie The Legend of Hell's Gate: An American Conspiracy and Joe Lynch's The Knights Of Badassdom, a RPG comedy about a group of LARPers. She also starred in the Sony Crackle exclusive drama series, Sequestered in 2014. Her latest projects are a minor character (Miss Jones) in the upcoming Netflix series Wu Assassins, which was released on August 2019 and a narration role in the sci-fi serial drama Alternis by Serial Box.

Summer Glau received a 2005 Saturn Award for Best Supporting Actress for her performance in Serenity.
In 2008 she received a Saturn Award for Best Supporting Actress for her performance as Cameron Phillips in Terminator: The Sarah Connor Chronicles.

Here is my long overdue thank you note to anyone and every person who has watched our pride and joy Terminator: The Sarah Connor Chronicles… Now that our season is done and I somehow feel far away, I want to say that it wouldn’t have been possible without faithful viewers; and it wouldn’t have been fun to make without such sincere interest from you…
